Cell fractionation is the process where cells are broken up and the different organelles are separated out.
The tissue is placed in a cold, isotonic buffered solution.
Cold - Reduces enzyme activity that could otherwise break down the organelles.
Isotonic - Prevents organelles from bursting or shrinking as a result of osmotic gain or loss of water because the water potential is the same as the cell.
Buffered - Maintains a constant pH.
